1. Introduction:
2-CTC resin is a highly acid-labile resin suitable for the preparation of peptides using the Fmoc strategy, as well as for the synthesis of protected peptide fragments. This resin is particularly well suited for preparing peptides containing C-terminal residues such as cysteine (Cys), histidine (His), methionine (Met), tyrosine (Tyr), and proline (Pro).

Cleavage of protected peptides from this support can be performed using a mixture of acetic acid (AcOH), trifluoroethanol (TFE), and dichloromethane (DCM), 0.5% trifluoroacetic acid (TFA), or hexafluoroisopropanol (HFIP). Alternatively, complete deprotection of peptides can also be achieved using the conventional 95% trifluoroacetic acid (TFA) cleavage method.

2. Main Functions:
2.1. Solid-phase synthesis support: 2-CTC resin serves as an acid-labile solid-phase support that covalently immobilizes substrates onto a polystyrene matrix via its chlorotrityl functional group, enabling efficient solid-phase reactions with simplified washing and purification.
2.2. Mild acid cleavage capability: Exhibits high acid sensitivity, enabling selective cleavage under mild conditions such as AcOH/TFE/DCM mixtures, low-concentration TFA, or HFIP, as well as complete deprotection under standard 95% TFA treatment.
2.3. Compatibility with Fmoc strategy: Fully compatible with the Fmoc-based solid-phase peptide synthesis strategy, enabling stepwise peptide elongation while maintaining the stability of side-chain protecting groups.
2.4. Improved selectivity and yield: Reduces side reactions, enhances coupling efficiency and product purity, and minimizes racemization and oligomerization byproducts.
2.5. Fragment-based synthesis capability: Supports fragment-based and modular synthesis strategies, enabling peptide fragment condensation and structural modification of complex peptides.
